Insomnia affects approximately 25% of adults. We have developed novel compounds with potent, selective, and orally active GPR50 agonists that modulate sleep. GPR50 is a 7-transmembrane G protein-coupled receptor expressed in multiple brain regions associated with sleep/wake behavior. We propose that GPR50 agonists provide a unique opportunity to modulate sleep via a novel mechanism of action without the need to rely on positive allosteric modulators of the GABA-B receptor. Our preliminary data indicates that GPR50 agonists bind the receptor with nanomolar affinity and have dose-dependent effects on sleep. Importantly, GPR50 agonists cause less sedation and brain excitation than positive allosteric modulators of the GABA-B receptor.
